Effect of chronic temperature exposure on the immunity of abalone, Haliotis discus hannai

AbstractThe effect of temperature on the immune response was studied in the abalone (Haliotis discus hannai) reared in seawater at 8, 14 and 26 degrees C for 30days, with abalone maintained at optimum temperature (20 degrees C) serving as the control. The total haemocyte count (THC) in abalone in the 8 degrees C treatment group was significantly higher than in the control group (P<0.05). A significantly (P<0.05) lower phagocytic capacity was observed in the 8 and 14 degrees C treatment groups. Reactive oxygen species (ROS) production in haemocytes of abalone in the 8 degrees C treatment group was significantly (P<0.05) higher than that in the control group. Real-time RT-PCR analyses indicated that the highest level of serine protease inhibitor (Spi) expression was detected in the 26 degrees C treatment group (P<0.05), and cysteine proteases inhibitor (Cpi) was also significantly higher in the 8 and 26 degrees C exposure groups. The transcription of the four immune defence genes was induced in all groups after challenge with Vibrio anguillarum. These results indicated that not only the temperature but also the synergy of thermal and bacteria stimulus may affect the immune response of abalone.
