AbstractPortable, low-cost and quantitative analysis of pathogen at home and in the field remain a key challenge in medical diagnostics and food safety. Despite many years of research, very few such tools are available. Motivated by present clinical requirements, pocket-sized device, such as commercial glucose meter (CGM), have emerged. Taking advantage of the CGM, we develop an approach to quantify bacteria. Molecular recognition elements, such as aptamer, coupled with a signal amplification method based on invertase-promoted hydrolysis of sucrose for the sensitive and selective detection of bacteria. This paper aims to develop a portable device combined with enzyme-mediated enhancement for biological/chemical analyte detection. A linear relationship between the response and the logarithm of the Staphylococcus aureus concentration using this method was obtained for concentrations ranging from 1.2 x 10(5) to 1.2 x 10(8) cfu mL(-1), with a detection limit of 1.0 x 10(5) cfu mL(-1). This study demonstrates an approach for bacteria detection and also provides a powerful platform to monitor bacterial infection, thus rendering potential broad antibacterial applications and bacteria diagnosis.
